In Windows 10:
- left-click the Start button
- hover the mouse over the power button
- left-click the username at the top of the list
- left-click Lock

Apparently there isn't a clickable way to lock a Windows machine.

Win+L does the job with the keyboard, but with mouse only I don't think there is a way.
